High Fidelity: SLU Researcher Finds Keys to Genome

LOUIS - Maintaining the stability and the correct sequence of our genetic information is vital to the accurate transmission of our genetic code. However, in the course of replicating, our DNA frequently runs into roadblocks, arising from both internal and external sources, that threaten the fidelity of our genetic information. The accurate processing of these roadblocks is paramount to genome integrity. Defects in this process can lead to cancer, genetic problems and premature aging. In a research paper published in the Journal of Cell Biology, Alessandro Vindigni, Ph.D., professor in the Edward A. Doisy Department of Biochemistry and Molecular Biology at Saint Louis University, shares a discovery that explains how cells use a process called replication fork reversal in order to deal with these roadblocks and transmit accurate genetic data. Lesions in DNA can occur as often as 100,000 times per cell per day. They can be the result of normal metabolic activities, like free radicals, as well as exposure to environmental factors such as UV radiation, X-rays and chemical compounds. In their report appearing in PNAS Early Edition, the researchers describe a smartphone-based device that uses the kind of technology used to make holograms to collect detailed microscopic images for digital analysis of the molecular composition of cells and tissues. "The global burden of cancer, limited access to prompt pathology services in many regions and emerging cell profiling technologies increase the need for low-cost, portable and rapid diagnostic approaches that can be delivered at the point of care," says Cesar Castro, MD, of the MGH Cancer Center and Center for Systems Biology, co-lead author of the report. VTT has developed a drug test printed on paper. VTT used antibodies -- produced by methods of molecular biology -- as morphine sensing molecules when creating this printing technology-based morphine test. Using printing technology to manufacture rapid tests enables high production volumes and low production costs. A paper-based test enables a rapid analysis of whether a compound -- in this case, morphine -- is present in a given sample. Possible future applications of the developed test include drug testing at workplaces and in connection with traffic control. This method, developed by VTT Technical Research Centre of Finland Ltd, provides several advantages, such as high production volumes, low material costs and disposability as well as design freedom based on bendability and foldability of paper. "In 2010, we proved that the VTT method works in a hemoglobin assay. Through our continued development efforts, we wanted to confirm that the method also works in mass-production of more demanding tests. Morphine as a small-sized molecule places major requirements on the analytical performance of the test.
