, are multipotent stem cells that can differentiate into a variety of cell types. Cell types that MSCs have been shown to differentiate into in vitro or in vivo include osteoblasts, chondrocytes, myocytes, adipocytes, and, as described lately, beta-pancreatic islets cells. MSCs are rare in bone marrow, representing ~1 in 10,000 nucleated cells. Although not immortal, they have the ability to expand manyfold in culture while retaining their growth and multilineage potential. MSCs are identified by the expression of many molecules including CD105 (SH2) and CD73 (SH3/4) and are negative for the hematopoietic markers CD34, CD45, and CD14. The properties of MSCs make these cells potentially ideal candidates for tissue engineering. It has been shown that MSCs, when transplanted systemically, are able to migrate to sites of injury in animals, suggesting that MSCs possess migratory capacity. However, the mechanisms underlying the migration of these cells remain unclear. Chemokine receptors and their ligands and adhesion molecules play an important role in tissue-specific homing of leukocytes and have also been implicated in trafficking of hematopoietic precursors into and through tissue. Vancouver, British Columbia (PRWEB) May 12, 2014 STEMCELL Technologies Inc. has just released NEW MesenCult Proliferation Kit with MesenPure (Mouse), a novel cell culture medium to advance research on mouse mesenchymal stem and progenitor cells (MSCs). When added to MesenCult medium, MesenPure supplement enriches mouse bone marrow- or compact bone-derived MSC cultures by reducing the number of hematopoietic cells. Culturing with MesenPure eliminates the time-intensive serial passaging steps and frequent cell culture medium changes normally required to decrease the unwanted hematopoietic cell population typically present in MSC cultures. Cultures treated with MesenPure appear homogeneous and mostly devoid of hematopoietic cells as early as passage zero and also contain increased numbers of mesenchymal stem cells that display more robust differentiation. Abstract Introduction Bone marrow mesenchymal stem cells (BMMSCs) are a heterogeneous population of postnatal precursor cells with the capacity of adhering to culture dishes generating colony-forming unit-fibroblasts (CFU-F). Here we identify a new subset of BMMSCs that fail to adhere to plastic culture dishes and remain in culture suspension (S-BMMSCs). To catch S-BMMSCs, we used BMMSCs-produced extracellular cell matrix (ECM)-coated dishes. Isolated S-BMMSCs were analyzed by in vitro stem cell analysis approaches, including flow cytometry, inductive multiple differentiation, western blot and in vivo implantation to assess the bone regeneration ability of S-BMMSCs. Furthermore, we performed systemic S-BMMSCs transplantation to treat systemic lupus erythematosus (SLE)-like MRL/lpr mice. S-BMMSCs are capable of adhering to ECM-coated dishes and showing mesenchymal stem cell characteristics with distinction from hematopoietic cells as evidenced by co-expression of CD73 or Oct-4 with CD34, forming a single colony cluster on ECM, and failure to differentiate into hematopoietic cell lineage. Moreover, we found that culture-expanded S-BMMSCs exhibited significantly increased immunomodulatory capacities in vitro and an efficacious treatment for SLE-like MRL/lpr mice by rebalancing regulatory T cells (Tregs) and T helper 17 cells (Th17) through high NO production. Dont be fooled by quacks and fake meds

Beware of fake medicines and advertisements touting the purported miracles that stem cell therapy can do. This was the warning aired by former health secretary Esperanza Cabral at the Kapihan sa Manila at the Diamond Hotel last Monday. Contrary to what the ads claim, she said, stem cell therapy has not been scientifically proven to cure any disease or make anyone young again. It has been successful in a very few experiments, which is the reason quack doctors are taking advantage of it to make exaggerated claims that the therapy can cure the deadliest diseases known to man. The Food and Drug Administration (FDA) very recently issued a similar warning against it. Stem cell therapy is the process of injecting into patients young cells taken from humans or sheep. The theory is that the young cells will rejuvenate the old cells of the patients and make them young again and cure whatever diseases they have. Although experiments are being conducted, no such results have been achieved. But that does not prevent foreign quack doctors from coming here and making all those exaggerated claims. Author information Abstract With technological advances in basic research, the intricate mechanism of secondary delayed spinal cord injury (SCI) continues to unravel at a rapid pace. However, despite our deeper understanding of the molecular changes occurring after initial insult to the spinal cord, the cure for paralysis remains elusive. Current treatment of SCI is limited to early administration of high dose steroids to mitigate the harmful effect of cord edema that occurs after SCI and to reduce the cascade of secondary delayed SCI. Recent evident-based clinical studies have cast doubt on the clinical benefit of steroids in SCI and intense focus on stem cell-based therapy has yielded some encouraging results. An array of mesenchymal stem cells (MSCs) from various sources with novel and promising strategies are being developed to improve function after SCI. In this review, we briefly discuss the pathophysiology of spinal cord injuries and characteristics and the potential sources of MSCs that can be used in the treatment of SCI.
