Ibrutinib and Hypertension in CLL: What Are the Risks? – Cancer Therapy Advisor

Posted: Published on November 23rd, 2019

Ibrutinib is an irreversible inhibitor of Bruton tyrosine kinase (BTK), which is an integral component of the B-cell receptor (BCR) and cytokine (CYK) pathways. Ibrutinib is approved by the US Food and Drug Administration for use in multiple B-cell related conditions including mantle cell lymphoma (MCL), chronic lymphocytic leukemia (CLL), small lymphocytic lymphoma (SLL), Waldenstrm macroglobulinemia (WM), marginal zone lymphoma (MZL), and chronic graft versus host disease (cGVHD).1 Ibrutinib use has been associated with multiple adverse events (AEs), of which cardiovascular (CV) toxicity represents one of the most commonly studied. The exact pathophysiology to explain the CV risks associated with ibrutinib have not yet been fully elucidated, although several mechanisms have been proposed Continue reading

Effects of genetic transfection on calcium cycling pathways mediated by double-stranded adeno-associated virus in postinfarction remodeling – DocWire…

Posted: Published on November 6th, 2019

OBJECTIVE: Restoring calcium sensor protein (S100A1) activity in failing heartsposes a promising therapeutic strategy. We hypothesize that cardiac overexpression of the S100A1 gene mediated by a double-stranded adeno-associated virus (scAAV) results in better functional and molecular improvements compared with the single-stranded virus (ssAAV). Heart failure was induced by coronary artery ligation. Continue reading

PH Common in Myeloproliferative Neoplasms, Linked to Diastolic Dysfunction – The Cardiology Advisor

Posted: Published on October 26th, 2019

Pulmonary hypertension (PH) is common among patients with myeloproliferative neoplasm (MPN) and is linked to left ventricular (LV) hypertrophy and diastolic dysfunction, according to study results published in the Journal of the American Society of Echocardiography. In this retrospective cohort study using consecutive patients with MPN who underwent transthoracic echocardiography between February 2011 and October 2017 (N=301), researchers used established cutoffs for PH (35 mm Hg) and advanced PH (50 mm Hg) and assessed LV and right ventricular (RV) performance via conventional indices. For patients with multiple echocardiographic examinations, they analyzed the initial examination that demonstrated impaired RV function to assess prognostic impact. Continue reading
