FDA grants orphan drug status for Targazyme's TZ101 for use with regulatory T cells

PBR Staff Writer Published 11 February 2015 The US Food and Drug Administration (FDA) has granted Orphan Drug designation to Targazyme's fucosyltransferase enzymes and small molecule product, TZ101, to prevent and reduce the severity and incidence of graft versus host disease (GVHD) in patients eligible for hematologic stem cell transplant. The company said that TZ101 could potentially transform hematopoietic stem cell transplantation by reducing patient morbidity and mortality from GVHD. GVHD is a serious, life-threating complication of stem cell transplantation and is very difficult to manage clinically. Targazyme chairman and chief executive officer Lynnet Koh said: "The granting of Orphan Drug status for TZ101 for prevention of GVHD in stem cell transplant patients, as well as our previous Orphan Drug designation of TZ101 for cord blood transplantation, provides additional validation of our innovative platform technologies. "TZ101 and our second product, TZ102 are enabling technologies for improving efficacy outcomes for multiple cell-based therapeutic approaches used to prevent and treat a variety of different diseases for which there is a high unmet medical need. Donor telomere length and survival after hematopoietic cell transplantation in patients with severe aplastic anemia

NCI News Note A new NCI study has found that, among patients with severe aplastic anemia (SAA) who received a hematopoietic cell transplant (HCT) from an unrelated donor, those whose donor white blood cells had longer telomeres had higher survival rates five-years after transplantation than those whose donor white blood cells had shorter telomeres. By contrast, the length of telomeres in a patients own white blood cells, as measured before transplantation, was not associated with survival. SAA, a disease in which the bone marrow fails to produce enough blood cells, can occur at any age but is most common in young adults; in HCT, donor blood or marrow stem cells, which are cells that give rise to other blood cells, are given to patients by intravenous infusion. Telomeres are complex structures on the ends of chromosomes that help maintain chromosome integrity. They shorten naturally as people age. Telomere length has been implicated in the development of several cancers, as well as other health conditions. The award fosters advancements in the field of stem cells and regenerative medicine by honoring a young researcher who is principle author of an article published in SCTM over the course of a year that is deemed to have the most impact and to push the boundaries of novel and insightful research. Dr. Dahlkes paper describes the discovery of a universal stem cell product that not only seems to increase the long-term survival of organ transplants in instances when the donor is not related to the recipient, but also retains that immunological privileged state when the organ is then transplanted into yet another unrelated recipient. The paper was published in the August 2013 issue of SCTM. And yet, one year ago, when 17-year-old Ankit was in need of a stem cell transplant to treat his acute myeloid leukaemia, 24-year-old Jinal donated her cells to him. It wasnt an easy process, said Raghu Rajagopal, chief executive officer of DATRI Blood Stem Cell Donors Registry, which brought the two together. On Wednesday, speaking at Apollo Hospitals, where the stem cell transplantation took place, he said getting the donation required counselling Jinals family, her fianc and his family, and even their community. But Jinal was determined to donate and made sure the process went through, he said. Over 1,00,000 Indians are diagnosed with blood cancer and other blood-related disorders every year. Stem cell donations can go a long way towards increasing their survival chances, said paediatric hemato-oncologist Revathi Raj. But finding donors within the family is very difficult, especially now, with smaller families. More people should come forward and register as donors. The family of a seven-year-old girl who was given just a 40 per cent chance of survival say clinical trials helped save her life. And they are now backing a national Cancer Research UK campaign highlighting the importance of the trials. Isabella Lyttle, from Shelfield, Walsall, took part in an immunotherapy trial to help her fight a rare type of cancer known as neuroblastoma when she was just three. Isabellas parents, Mark and Jennie, want people to see clinical trials as a normal and essential part of research and treatment for cancer. The trials show whether new tests and treatments are safe, what their side-effects are and whether they are better than what is currently used. Mark said: We know from personal experience that clinical trials are vital, both to people like Isabella who received treatment and to future generations. Today, more and more people are surviving thanks to research, and cancer trials are crucial in helping to develop better and kinder treatments. Isabella, a pupil at St Francis Catholic Primary School in Shelfield, was diagnosed with cancer in October 2010 after complaining of a pain in her hip.
