Challenges in Gene Therapy – Learn Genetics

Posted: Published on November 4th, 2015

HOME Gene Therapy Challenges in Gene Therapy? Gene therapy is not a new field; it has been evolving for decades. Despite the best efforts of researchers around the world, however, gene therapy has seen only limited success. Why? Gene therapy poses one of the greatest technical challenges in modern medicine. It is very hard to introduce new genes into cells of the body and keep them working. And there are financial concerns: Can a company profit from developing a gene therapy to treat a rare disorder? If not, who will develop and pay for these life-saving treatments? Let's look at some of the main challenges in gene therapy. For some disorders, gene therapy will work only if we can deliver a normal gene to a large number of cellssay several millionin a tissue. And they have to the correct cells, in the correct tissue. Once the gene reaches its destination, it must be activated, or turned on, to make the protein it encodes. And once it's turned on, it must remain on; cells have a habit of shutting down genes that are too active or exhibiting other unusual behaviors. Introducing changes into the wrong cells Targeting a gene to the … Continue reading

Woman's genetic counselors can help you understand your genetic risks for certain diseases, such as cancer, or for passing an existing disease on to a child. Genetic counseling can lead to the earliest detection of diseases you or your baby may be at-risk of developing. If you are concerned about diseases that run in your family, talk to you doctor about genetic counseling. Genetics is the study of heredity, the process in which parents pass certaingenesonto their children. A person's physical appearance height, hair color, skin color and eye color are determined by genes. Other characteristics affected by heredity include: Humans have an estimated 100,000 different genes that contain specific genetic information, and these genes are located on stick-like structures in the nucleus of cells called chromosomes. When a gene is abnormal, or when entire chromosomes are left off or duplicated, defects in the structure or function of the body's organs or systems can occur. These mutations or abnormalities can result in disorders such as cystic fibrosis, a recessive genetic disease, or Down syndrome, an abnormality that occurs when a baby receives three No. 21 chromosomes. Each person has more than 100,000 genes that direct the growth and development of … Continue reading

Y.W. Kans pioneering research into the hemoglobinopathies sickle cell anemia and thalassemia has widely impacted genetic research, diagnostics, and treatment of human disease. The Institute for Human Genetics is proud to recognize Y.W. Kan with a symposium honoring his decades-long contributions. Y.W. Kan arrived at UCSF in the 1970s when he and many others (including Herb Boyer and Bishop & Varmus) helped usher in the era of molecular genetics. With long-time collaborator Andre Dozy, he discovered the first polymorphism in human DNA by Southern blot analysis in 1978, launching the ability to map genes on human chromosomes. He and another long-time collaborator, Judy Chang, used those same techniques in 1979 to show how missing genes cause disease. He is the recipient of many national and international awards for his contributions. He continues to investigate the treatment of these diseases using stem cell and iPS cell therapies. The Symposium will feature presentations from James Gusella, Katherine High, Dennis Lo, Bertram Lubin, Robert Nussbaum, Stuart Orkin, and Griffin Rodgers. Stuart Orkin will be featured as the 2015 Charles J. and Lois B. Epstein Visiting Professor. Featured topics will includegene mapping, gene therapy, hemoglobinopathies, and non-invasive prenatal testing. The IHG Symposium will be … Continue reading

Genetics – Simple English Wikipedia, the free encyclopedia

Genetics is a discipline of biology.[1] It is the science of heredity. This includes the study of genes, and the inheritance of variation and traits of living organisms.[2][3][4] In the laboratory, genetics proceeds by mating carefully selected organisms, and analysing their offspring. More informally, genetics is the study of how parents pass some of their characteristics to their children. It is an important part of biology, and gives the basic rules on which evolution acts. The fact that living things inherit traits from their parents has been known since prehistoric times, and used to improve crop plants and animals through selective breeding. However, the modern science of genetics, which seeks to understand the process of inheritance, only began with the work of Gregor Mendel in the mid-nineteenth century.[5] Although he did not know the physical basis for heredity, Mendel observed that organisms inherit traits via discrete units of inheritance, which are now called genes. Living things are made of millions of tiny self-contained components called cells. Inside of each cell are long and complex molecules called DNA.[6]DNA stores information that tells the cells how to create that living thing. Parts of this information that tell how to make one small … Continue reading

CAR T-Cell Immunotherapy for ALL – National Cancer Institute

On This Page For years, the cornerstones of cancer treatment have been surgery, chemotherapy, and radiation therapy. Over the last decade, targeted therapies like imatinib (Gleevec) and trastuzumab (Herceptin)drugs that target cancer cells by homing in on specific molecular changes seen primarily in those cellshave also emerged as standard treatments for a number of cancers. Illustration of the components of second- and third-generation chimeric antigen receptor T cells. (Adapted by permission from the American Association for Cancer Research: Lee, DW et al. The Future Is Now: Chimeric Antigen Receptors as New Targeted Therapies for Childhood Cancer. Clin Cancer Res; 2012;18(10); 278090. doi:10.1158/1078-0432.CCR-11-1920) And now, despite years of starts and stutter steps, excitement is growing for immunotherapytherapies that harness the power of a patients immune system to combat their disease, or what some in the research community are calling the fifth pillar of cancer treatment. One approach to immunotherapy involves engineering patients own immune cells to recognize and attack their tumors. And although this approach, called adoptive cell transfer (ACT), has been restricted to small clinical trials so far, treatments using these engineered immune cells have generated some remarkable responses in patients with advanced cancer. For example, in several early-stage trials … Continue reading

Understanding Stem Cell Transplant VICC Momentum

Vanderbilt-Ingram Cancer Center performs around 215 stem cell and bone marrow transplants each year, providing care leading up to the transplant, through all aspects of the procedure, and indefinitely after the transplant to monitor for complications. To address some common questions about the procedure, we spoke with Madan Jagasia, M.D., director of the Outpatient Transplant Program and section chief for Hematology and Stem Cell Transplant at Vanderbilt-Ingram. What is a stem cell? (click to enlarge) Stem cells or more specifically, in this case, hematopoietic stem cells are cells that can give rise to all the different types of mature blood cells the red blood cells that carry oxygen, the platelets involved in blood clotting, and a host of white blood cells, which are part of the bodys immune system and provide defense against infectious agents. Stem cells are self-renewing (i.e., they can produce more of themselves) and reside primarily in the bone marrow but also circulate in the blood. For a stem cell transplant, the stem cells can come from a related or unrelated donor, from umbilical cord blood, or from the patient him/herself. No, these are adult stem cells, from the blood or bone marrow. Even when the source … Continue reading
