Maarten Holl EVIDENCE: Susan Vintiner, an ESR scientist, gave evidence about the results of DNA samples taken in the Lundy case. Paint found on items at the scene of the alleged Lundy murders could have matched the paint Mark Lundy used on his tools. Lundy, 56, is accused of staging a burglary, and killing wife, Christine, 38, and daughter Amber, 7 on August 30, 2000. The Crown alleges the pair were killed with a small axe or tomahawk, which has not been found. Lundy was tried in 2002 and is being retried following a Privy Council ruling. He has pleaded not guilty. Our reporter Jono Galuszka was at court in Wellington and updated developments throughout the day. KEY POINTS: *Tests done on samples taken from Mark Lundy's shirt found DNA which was extremely likely to have come from Christine Lundy, the jury heard. *One of the samples from Lundy's polo shirt contained unknown female DNA, the court has heard. *A third American pathologist has told the court tissue found on Mark Lundy's shirt was central nervous system tissue. Using single cell transcriptogenomics to probe the cell's defense mechanisms, study published in Mutation Research -- Fundamental and Molecular Mechanisms of Mutagenesis The central dogma of molecular biology describes the flow of genetic information. It was first described by Francis Crick in 1956 as one-way traffic: as: "DNA makes RNA and RNA makes protein." A recent paper published in Mutation Research - Fundamental and Molecular Mechanisms of Mutagenesis, however suggests that, rather than being a one-way street, DNA-directed RNA transcription may have profound adaptability. The authors of the paper showed a conceptually novel relationship between the genotype (DNA) and the phenotype (the products of the transcription of DNA). The method the authors used to make this discovery is termed Single-Cell Transcriptogenomics (SCTG). It allows DNA and RNA sequencing to be performed concurrently on the same single cells taken from a cell population treated with the powerful mutagen ethylnitrosourea. This method allowed the authors, for the first time, to prove the tendency of the transcriptional machinery in the cell to avoid transcribing DNA strands harboring a newly induced mutation. Murray Close/Getty Human mutations are not just a favourite device in science-fiction films such as the X-men series, they also offer a way to mark time in the evolutionary past. Mathematicians keep refining even though they know it to more than 12trillion digits; physicists beat themselves up because they cannot pin down the gravitational constant beyond three significant figures. Geneticists, by contrast, are having trouble deciding between one measure of how fast human DNA mutates and another that is half that rate. The rate is key to calibrating the molecular clock that puts DNA-based dates on events in evolutionary history. So at an intimate meeting in Leipzig, Germany, on 2527 February, a dozen speakers puzzled over why calculations of the rate at which sequence changes pop up in human DNA have been so much lower in recent years than previously. They also pondered why the rate seems to fluctuate over time. The meeting drew not only evolutionary geneticists, but also researchers with an interest in cancer and reproductive biology fields in which mutations have a central role.
