Engineers put the 'squeeze' on human stem cells

Forces applied to the bead cause ion channels in the ER to open mechanically (shown... view more Credit: Jie Sun/UC San Diego After using optical tweezers to squeeze a tiny bead attached to the outside of a human stem cell, researchers now know how mechanical forces can trigger a key signaling pathway in the cells. The squeeze helps to release calcium ions stored inside the cells and opens up channels in the cell membrane that allow the ions to flow into the cells, according to the study led by University of California, San Diego bioengineer Yingxiao Wang. Researchers have known that mechanical forces exerted on stem cells have a significant role to play in how the cells produce all kinds of tissues--from bone to blood--from scratch. But until now, it hasn't been clear how some of these forces translate into the signals that prod the stem cells into building new tissue. The findings published in the journal eLife could help scientists learn more about "the functional mechanisms behind stem cell differentiation," said Wang, an associate professor of bioengineering. They may also guide researchers as they … Continue reading

Infants with rare bone disease improve bone formation after cell transplantation

Posted: Published on March 10th, 2015

Severe hypophosphatasia generally fatal during infancy, bone marrow transplant along with mensenchymal stem cell transplants offers hope Putnam Valley, NY. (Feb 9, 2015) - Recent research carried out by a team of researchers in Japan has investigated the use of bone marrow transplants (BMTs) to treat hypophosphatasia (HPP). In this study, the researchers carried out BMT for two infants with HPP in combination with allogenic (other-donated) mesenchymal stem cell transplants (MSCTs). The allogenic MSC donors were a parent of the infant. The study will be published in a future issue of Cell Transplantation and is currently freely available on-line as an unedited early e-pub at: http://ingentaconnect.com/content/cog/ct/pre-prints/content-CT-1337_Taketani_et_al "Hypophosphatasia" (HPP) is a rare and most often fatal genetic bone disease affecting infants that has no current treatment. The disease is caused by mutations in the ALPL gene, which encodes alkaline phosphatase (ALP). Patients with severe HPP develop bone impairment and have extremely low levels of ALP activity, an enzyme necessary for bone mineralization. Although there are mild and more severe forms, severe hypophosphatasia prevents proper bone mineralization during perinatal development. When the disease develops perinatally, many infants are still-born, with little evidence of bone mineralization. HPP can also appear in later infancy, … Continue reading

IVF test improves chances of implantation by pinpointing fertility window

Posted: Published on March 10th, 2015

The scientists behind the technique believe IVF frequently fails because the embryo is transferred at the wrong time. Photograph: Alamy Thousands of infertile couples could benefit from a new test that tailors the timing of IVF treatment to a womans individual cycle for the first time. The scientists behind the technique believe that IVF frequently fails because the embryo is transferred at the wrong time, missing a crucial fertility window. The new test assesses the activity of genes of the womb lining to pinpoint a womans optimum time for treatment and in pilot studies the personalised approach appeared to significantly boost success rates. Prof Juan Garcia-Velasco, of the IVI fertility clinic in Madrid, said: We think that about 15% of cases of implantation failure are simply due to bad timing. Prof Garcia-Velasco is now leading a clinical trial of the test, involving 2,500 patients in more than ten countries, including Britain. Geeta Nargund, medical director of Create Fertility whose London clinic is participating, said: The weakest link in IVF is implantation failure. I believe this is a breakthrough. There are more than 60,000 IVF cycles in Britain each year, but just 24% of these treatments lead to live births. Clinics … Continue reading
