Optimism on stem cells, ahead of reality

And as the level of desperation rises, some patients will pay over the odds for treatment, pursing unproven options in the hope of some improvement in their condition. And where there is unmet demand, supply soon steps in to fill the gap. Last year, there was intense global media attention on stem cell treatments following a paralysed patient in Poland who walked after a cell transplant, a project involving Polish and UK researchers. Stem cells may well offer significant potential promise for patients in a range of treatments. But to date, much of that optimism has run well ahead of the reality. Stem cell treatment has California researchers a step closer to HIV cure

DAVIS, CA (KOVR) - Researchers at UC Davis say they are one step closer to finding a cure for HIV in a breakthrough study for millions around the world living with the virus. At 60 years old, Paul Curtis looks like the picture of health. I exercise, eat well get a lot of rest, he said. But 30 years ago, Curtis was diagnosed as HIV-positive. Doctors told him he might have a year to live, but he's proven them wrong. With this disease, it's imperative that you take the medications consistently, Curtis said. He relies on medication daily. At one point he took more than 40 pills a day. And he can't miss a dose. The virus mutates rapidly when you miss doses, he said. He's one of millions worldwide waiting for a cure. Previous studies have come close, but none have proven to fight off the virus with stem cell therapy. Dr. Joe Anderson says he has developed genetically modified human stem cells, which have resisted infection in mice. Abstract Introduction Different mesenchymal stromal cells (MSC) have been successfully isolated and expanded in vitro and nowadays they are tested in clinical trials for a wide variety of diseases. Whether all MSC express the same cell surface markers or have a similar secretion profile is still controversial, making it difficult to decide which stromal cell may be better for a particular application. We isolated human mesenchymal stromal cells from bone marrow (BM), adipose tissue (AT) and Whartons jelly (WJ) and cultured them in fetal bovine serum supplemented media. We evaluated proliferation, in vitro differentiation (osteogenic, adipogenic and chondrogenic potential), expression of cell surface markers and protein secretion using Luminex and ELISA assays. Cell proliferation was higher for WJ-MSC, followed by AT-MSC. Differences in surface expression markers were observed only for CD54 and CD146. WJ-MSC secreted higher concentrations of chemokines, pro-inflammatory proteins and growth factors. AT-MSC showed a better pro-angiogenic profile and secreted higher amounts of extracellular matrix components and metalloproteinases. Mesenchymal stromal cells purified from different tissues have different angiogenic, inflammatory and matrix remodeling potential properties. The Manipal University unit of the UNESCO Chair in Bioethics, in conjunction with the Department of Obstetrics and Gynaecology and the Division of Clinical Embryology, KMC, Manipal, conducted a national seminar on Assisted Reproductive Technology Challenges, concerns and ethics here recently. The lectures were delivered by speakers which included Vijay Kumar Bhujang (United States), Russell DSouza (Australia), Anand Ramakrishna (United Kingdom) and Shobana Mohandas (India). A panel discussion on Sharing Experiences in Assisted Reproductive Technology was held. Mr. DSouza, Asia Pacific Chair of the UNESCO Chair in Bioethics, introduced the ethical issues that arise in medically assisted reproduction with regards to the biological parents, the surrogate parent and the child. Mr. Ramakrishna, from the United Kingdom, stressed on the evolution of various legislation regarding the right of the conceived child to know the identity of the parent.
