Seamless gene correction of beta-thalassemia mutations in patient-specific cells

A major hurdle in gene therapy is the efficient integration of a corrected gene into a patient's genome without mutating off-target sites. In a paper published today in Genome Research, scientists have used CRISPR/Cas genome editing technology to seamlessly and efficiently correct disease-causing mutations in cells from patients with -thalassemia. -thalassemia results from inherited DNA mutations in the hemoglobin beta (HBB) gene, resulting in reduced HBB expression in red blood cells and, in the most severe forms, anemia. The only established curative treatment is hematopoietic stem cell transplantation; however, this treatment requires a matched donor. Gene therapy, which delivers a corrected copy of a gene into patient cells, could bypass the need for a donor. Previous attempts using a virus to randomly insert a normal gene into the genome has been successful in one -thalassemia patient, but the long-term effect of viral insertion is not yet known. To correct HBB mutations directly in a patient's genome, researchers first generated induced pluripotent stem cells, or iPSCs, from skin cells of patients. An Embryonic Cell’s Fate Is Sealed by the Speed of a Signal

Contact Information Available for logged-in reporters only When embryonic cells get the signal to specialize the call can come quickly. Or it can arrive slowly. Now, new research from Rockefeller University suggests the speed at which a cell in an embryo receives that signal has an unexpected influence on that cells fate. Until now, only concentration of the chemical signals was thought to matter in determining if the cell would become, for example, muscle, skin, brain or bone. It turns out that if ramped up slowly enough an otherwise potent signal elicits no response from the receiving cells. Meanwhile, a pulsing, on-off signal appears to have a stronger effect than a constant one, says researcher Ali Brivanlou, Robert and Harriet Heilbrunn Professor and head of the Laboratory of Molecular Vertebrate Embryology. This research is the latest collaboration between Brivanlou and Eric Siggia, Viola Ward Brinning and Elbert Calhoun Brinning Professor at Rockefellers Center for Studies in Physics and Biology. The founder of Argentina's leading human rights group said Monday that she had located the grandson taken from her daughter while a prisoner of the military dictatorship in the 1970s, one of the long-unsolved mysteries from the "dirty war" era that still haunts the country. Surrounded by her large extended family, an emotional Estela Barnes de Carlotto, founder of the Grandmothers of the Plaza de Mayo, announced that her long hunt for her grandchild had ended, while acknowledging other families are still searching for hundreds of children taken under similar circumstances. "Thanks to God, thanks to life, because I didn't want to die without embracing him and soon I will be able to," the 83-year-old grandmother said at a news conference covered live on national TV. She has not yet met him.
