Biologists describe mechanism promoting multiple DNA mutations

Posted: Published on July 30th, 2014

PUBLIC RELEASE DATE: 30-Jul-2014 Contact: Gary Galluzzo gary-galluzzo@uiowa.edu 319-384-0009 University of Iowa DNA mutationslong known to fuel cancer as well as evolutionary changes in a living organismhad been thought to be rare events that occur randomly throughout the genome. However, recent studies have shown that cancer development frequently involves the formation of multiple mutations that arise simultaneously and in close proximity to each other. These groups of clustered mutations are frequently found in regions where chromosomal rearrangements take place. The discovery, published in the journal Cell Reports, may one day lead to new cancer therapies, according to a University of Iowa biologist and her colleagues, and a group of researchers from the National Institute of Environmental Health Sciences led by Senior Associate Scientist Dmitry Gordenin. The formation of clustered mutations may result from the process of DNA repair. Anna Malkova, associate professor of biology in the UI College of Liberal Arts and Sciences, notes that the DNA repair pathway, known as break-induced replication (BIR), can promote clusters of DNA mutations. "Previously, we have shown that double-strand DNA breaks, which can result from oxidation, ionizing radiation and replication errors, can be repaired by BIR," says Malkova. "During BIR, one broken DNA … Continue reading

Cancer biomarkers: Written in blood

Illustration by Oliver Munday In 2012, Charles Swanton was forced to confront one of cancer's dirtiest tricks. When he and his team at the Cancer Research UK London Research Institute sequenced DNA from a handful of kidney tumours, they expected to find a lot of different mutations, but the breadth of genetic diversity within even a single tumour shocked them. Cells from one end differed from those at the other and only one-third of the mutations were shared throughout the whole mass. Secondary tumours that had spread and taken root elsewhere in the patients' bodies were different again1. The results confirmed that the standard prognostic procedure for cancer, the tissue biopsy, is woefully inadequate like trying to gauge a nation's behaviour by surveying a single street. A biopsy could miss mutations just centimetres away that might radically change a person's chances for survival. And although biopsies can provide data about specific mutations that might make a tumour vulnerable to targeted therapies, that information is static and bound to become inaccurate as the cancer evolves. Swanton and his team laid bare a diversity that seemed insurmountable. I am still quite depressed about it, if I'm honest, he says. And if we … Continue reading

More DNA Testing Ordered in 2003 Rape, Burn, Attempted Homicide Case

Posted: Published on July 30th, 2014

Brown County - More than a decade after a Brown County woman was brutally attacked and left for dead, one of the men convicted is again trying to get the case reopened. Action 2 News has learned the Wisconsin Innocence Project is currently sending evidence to a private lab, trying to prove Juan Nieto was not involved. What happened in a rural area of Brown County in 2003 is so horrific, it's unimaginable to most people. Investigators say two men kidnapped a woman from outside a Green Bay bar, raped her and set her on fire.Somehow she survived and got help. More than a year later, investigators tracked down Gregario Morales and Juan Nieto. DNA linked Morales to the crime. He pleaded guilty and agreed to testify against Nieto as part of the deal. That helped a jury convict Nieto, even though none of his DNA was ever found on any evidence. He's always maintained innocence. "He has never indicated at any time that he is anything but completely innocent," Timothy Geary, Nieto's trial attorney told Action Two News after sentencing in November of 2005. Last year, Nieto's case took a turn when the Wisconsin Innocence Project convinced a judge … Continue reading

Apartment complex requires residents to DNA test dogs to keep area clean

Posted: Published on July 30th, 2014

DNA testing has gone to the dogs. One of the newest apartment complexes in the metro wants dog owners to pick up after their dogs and they're using DNA testing to make sure they do. At River Park Place on River Road, the complex says they are a pet-friendly environment. But they also say they are a people-friendly environment. They want it to be clean. So that's why if you're a resident there, and you have a dog, it will have to be DNA tested. As one of the newest condominium and apartment complexes on the river, River Park Place is trying to set the bar high for luxury living. So to fix the problem, management called Poo Prints of Louisville. We take DNA swabs of pets on certain properties so it's easier for the property to track people who aren't picking up after their pet, said Joseph Tucker with Poo Prints of Louisville. With the DNA test kit, every dog owner at the complex will have to swab the jowls of their dog and submit the sample. Each dog will be identified by a tag. If feces matching their DNA are found on the property, it's a $200 fine … Continue reading
