Stem cell breakthrough in treating heart attacks

Picture: Murry Lab/University of Washington/PA Stem cell heart repair treatments could be tested on human patients within four years following a ground-breaking study of monkeys. Scientists successfully restored damaged cardiac muscle in macaque monkeys suffering the after-effects of experimentally induced heart attacks, paving the way to clinical trials. Researchers injected 1bn immature heart muscle cells derived from human embryonic stem cells into each animals heart. Over several weeks, the new cells developed, assembled into muscle fibres, and began to beat in correct time. On average, 40% of the damaged heart tissue was regenerated. It is the first time stem cell therapy for damage caused by heart attacks has been shown to work in a primate. These induced pluripotent stem cells (iPSCs) produced sperm precursors following transplantation into the testes of mice. The findings help to explain a genetic cause of male infertility and offer a window into basic sperm biology. The approach also holds considerable potential for clinical application, the researchers say. "Our results are the first to offer an experimental model to study sperm development," said Renee Reijo Pera of the Institute for Stem Cell Biology & Regenerative Medicine and Montana State University. "Therefore, there is potential for applications to cell-based therapies in the clinic, for example, for the generation of higher quality and numbers of sperm in a dish. "It might even be possible to transplant stem-cell-derived germ cells directly into the testes of men with problems producing sperm," she added. Home > News > technology-news Washington, May 1 : In a pioneering work, researchers at Columbia Engineering have successfully grown fully functional human cartilage from human stem cells derived from bone marrow tissue. "We have been able to generate fully functional human cartilage from mesenchymal stem cells by mimicking in vitro the developmental process of mesenchymal condensation," explained Gordana Vunjak-Novakovic, the Mikati Foundation professor of biomedical engineering at Columbia Engineering. This could have clinical impact, as this cartilage can be used to repair a cartilage defect, or in combination with bone in a composite graft grown in lab for more complex tissue reconstruction, he informed. The general approach to cartilage tissue engineering has been to place cells into a hydrogel and culture them in the presence of nutrients and growth factors and sometimes also mechanical loading. But using this technique with adult human stem cells has invariably produced mechanically weak cartilage. So Vunjak-Novakovic and her team wondered if a method resembling the normal development of the skeleton could lead to a higher quality of cartilage. PUBLIC RELEASE DATE: 1-May-2014 Contact: Glenna Picton picton@bcm.edu 713-798-7973 Baylor College of Medicine HOUSTON (May 1, 2014) -- A team of researchers led by Baylor College of Medicine have identified the gene underlying a newly recognized genetic syndrome that has symptoms of sleep apnea, delayed speech and hyptonia, or generalized upper body weakness. The study published online today in the American Journal of Human Genetics. The Baylor researchers first studied a patient from Australia with these symptoms who had been seen by many doctors and had multiple diagnostic tests, without any diagnosis. Although there was no family history of the disease, the researchers performed DNA sequence analysis on the patient and her parents to determine if there was an underlying genetic cause for her symptoms. The results showed damaging mutations had newly arisen in five genes in the patient when compared with the parents DNA sequence. One gene was a candidate for causing the disease because similar mutations were never seen in healthy control individuals.
