Background Friedreich's ataxia is a progressive degenerative disorder caused by deficiency of the frataxin protein. Expanded GAA repeats within intron 1 of the frataxin (FXN) gene lead to its heterochromatinisation and transcriptional silencing. Preclinical studies have shown that the histone deacetylase inhibitor nicotinamide (vitamin B3) can remodel the pathological heterochromatin and upregulate expression of FXN. We aimed to assess the epigenetic and neurological effects and safety of high-dose nicotinamide in patients with Friedreich's ataxia. Nicotinamide was generally well tolerated; the main adverse event was nausea, which in most cases was mild, dose-related, and resolved spontaneously or after dose reduction, use of antinausea drugs, or both. Phase 1 showed a dose-response relation for proportional change in frataxin protein concentration from baseline to 8 h post-dose, which increased with increasing dose (p=00004). Bayesian analysis predicted that 38 g would result in a 15-times increase and 75 g in a doubling of frataxin protein concentration. A transgender woman serving a three-year sentence is suing the Ohio Department of Rehabilitation and Correction after being denied medically necessary hormone treatments. Whitney Lee A transgender inmate has filed a lawsuit against the Ohio Department of Rehabilitation and Correction after her hormone treatments were discontinued in February 2012. Whitney Lee, a transgender woman serving a three-year sentence at Mansfield Correctional Institution on forgery and theft charges, began hormone replacement therapy in 1999. Her treatment was halted in February 2012 after a prison psychiatrist concluded that Lee did not meet the medical criteria for gender dysphoria, and therefore should no be allowed continued access to treatment. Gender dysphoria is characterized in the latest edition of the American Psychiatric Association's Diagnostic and Statistical Manual of Mental Disorders as "a marked difference between the individual's expressed/experienced gender and the gender others would assign him or her." The DSM elaborates, "It must continue for at least six months. In children, the desire to be of the other gender must be present and verbalized. This condition causes clinically significant distress or impairment in social, occupational, or other important areas of functioning." Lee, 36, has been living as a woman since she was 18. Stem cells made from the skin of adult, infertile men yield primordial germ cells cells that normally become sperm when transplanted into the reproductive system of mice, according to researchers at the Stanford University School of Medicine and Montana State University. The infertile men in the study each had a type of genetic mutation that prevented them from making mature sperm a condition called azoospermia. The research suggests that the men with azoospermia may have had germ cells at some point in their early lives, but lost them as they matured to adulthood. Although the researchers were able to create primordial germ cells from the infertile men, their stem cells made far fewer of these sperm progenitors than did stem cells from men without the mutations. The research provides a useful, much-needed model to study the earliest steps of human reproduction. Researchers at University of Adelaide have discovered that stem cells taken from teeth can grow to resemble brain cells and can be used in the brain as a therapy for stroke. "Stem cells from teeth have great potential to grow into new brain or nerve cells and this could potentially assist with treatments of brain disorders, such as stroke," explained Kylie Ellis, a commercial development manager with the University's Adelaide Research & Innovation (ARI). In the University's centre for stem cell research, lab studies have shown that stem cells from teeth can develop and form complex networks of brain-like cells.
