Stem Cell Survival Strategy Is Key to Blood and Immune System Health

Stem cells of the aging bone marrow recycle their own molecules to survive and keep replenishing the blood and immune systems as the body ages, researchers at UC San Francisco (UCSF) have discovered. The recycling process, known as autophagy, or self-eating, involves reusing molecules and the chemical energy obtained from these molecules to withstand the killing effect of metabolic stress that intensifies as the body ages. Emmanuelle Passegu, PhD The discovery, reported online Feb. 6 in the journal Nature, showed that autophagy allows stem cells to avoid the alternative response to stress, which is programmed cellular suicide, in which cells that arent up to snuff kill themselves for the greater good. While this trick of autophagy may help delay the onset of anemia, immune-system failure and other maladies that occur with age, as a survival strategy it is a bit of a compromise, said the senior author of the study, Emmanuelle Passegu, PhD, of the Eli and Edythe Broad Center of Regeneration Medicine and Stem Cell Research at UCSF. The study used a patient's skin cells to create motor neurons - nerve cells that control muscle activity - and the cells that support them called astrocytes. Researchers studied these two types of cells in the laboratory. They found that a protein expressed by abnormalities in a gene linked to motor neurone disease, which is called TDP-43, caused the astrocytes to die. The study, led by the University of Edinburgh and funded by the Motor Neurone Disease Association, provides fresh insight into the mechanisms involved in the disease. Although TDP-43 mutations are a rare cause of motor neurone disease (MND), scientists are especially interested in the gene because in the vast majority of MND patients, TDP-43 protein (made by the TDP-43 gene) forms pathological clumps inside motor neurons. Motor neurons die in MND leading to paralysis and early death. This study shows for the first time that abnormal TDP-43 protein causes death of astrocytes. Feb. 11, 2013 Using cutting-edge time-lapse photography, Keck School of Medicine researchers have discovered clues to the development of the head at the cellular level, which could point scientists to a better understanding of how organs and birth defects form in humans. A team of researchers at the Eli and Edythe Broad Center for Regenerative Medicine and Stem Cell Research at USC has for the first time determined the role of two important molecular signaling pathways that help control the number and position of repeated units of cells that pattern the head and face. Two members of a "Wnt" signaling pathway are instrumental in forming pharyngeal pouches that organize the structure of the head and face. Problems with forming the pouches can result in birth defects, including the rare DiGeorge syndrome, which causes an array of symptoms including an abnormal facial appearance, cleft palate, congenital heart disease, and loss of the thyroid and thymus. The research, "Wnt-Dependent Epithelial Transitions Drive Pharyngeal Pouch Formation," was published Feb. 11, 2013 in Developmental Cell.
