Umbilical cord cells outperform bone marrow cells in repairing damaged hearts

Posted: Published on November 16th, 2012

Public release date: 15-Nov-2012 [ | E-mail | Share ] Contact: Erin Vollick comm.ibbme@utoronto.ca 416-946-8019 University of Toronto TORONTO, Ontario (13 November, 2012) - A study published this month by researchers at the University of Toronto and Toronto's Princess Margaret Hospital has shown that cells derived from the umbilical cord, "Human Umbilical Cord PeriVascular Cells" (HUCPVCs), are more effective in restoring heart function after an acute myocardial infarction (in common parlance, a heart attack) in a pre-clinical model than a similar cell population derived from bone marrow. At present, mesenchymal cells, known to release a series of factors that stimulate tissue repair, and control inflammation, are most commonly harvested from bone marrow. But the new study, headed by Dr. Armand Keating, now suggests that umbilical cord cells outperform bone marrow cells in improving heart muscle function. The study, released in Cell Transplantation this month, demonstrates that the cells originating from the tissues surrounding the blood vessels of the human umbilical cord, also known as "Wharton's Jelly," outperformed the current gold standard for stem cell therapies for repairing damage to heart muscles, after an induced heart attack when injected directly into the affected area. Dr. Keating calls the HUCPVC results "statistically … Continue reading

Human umbilical cord blood cell co-culture supports embryonic stem cell expansion

Posted: Published on November 16th, 2012

Researchers in Taiwan have developed a "safe, feasible and robust co-culture system" supplied by human umbilical cord mensenchymal stem cells (HUCMSCs) to feed the sustained culture used for human embryonic stem cell (hESC) expansion prior to cell transplantation. The co-culture, said the researchers, "appears to eliminate the most feared characteristic of transplanted hESCs," which is their propensity to form tumors. The study, published in the current issue of CELL TRANSPLANTATION, is now freely available on-line at http://www.ingentaconnect.com/content/cog/ct/. "Embryonic stem cells have pluripotent potential," said study co-author Dr. Tang-Yuan Chu of the Buddhist Tzu Chi General Hospital at Tzu Chi University in Hualien, Taiwan. "The sustained maintenance of hESCs depends on a co-culture with an animal based "feeder" that can create the risk for transmitting nonhuman materials and unknown pathogens. To solve this problem, human tissues have been used as feeders." The expansion of pluripotent hESCs traditionally requires a feeder culture, meaning that a variety of animal and human tissues have been used in feeder cultures. The researchers note that while hESCs have been successfully co-cultured using human fetal muscle and skin cells, adult fallopian tube epithelial cells, foreskin cells, and bone marrow stem cells, their study used hUCMScs to create … Continue reading

Study reveals insights that could aid in therapeutic use of mesenchymal stem cells

Posted: Published on November 16th, 2012

ScienceDaily (Nov. 15, 2012) Mesenchymal stem cells (MSCs), are a newly emerging cellular therapy being tested in approximately 250 clinical trials worldwide to help repair damaged tissues, such as injured heart muscle following a heart attack. The problem is that when culture-expanded MSCs are injected into the circulation, they have trouble gaining access to the inflamed tissues -- exactly where their help is needed. Now, research led by investigators at Beth Israel Deaconess Medical Center (BIDMC) and Brigham and Women's Hospital (BWH) reveals new insights into how MSCs "traffic" from the circulation into the tissue, providing important clues that could be used to improve the delivery of this promising therapy. The findings are published in the November issue of the journal Stem Cells. MSCs have great clinical potential due to their convenient isolation, their lack of significant immunogenicity (allowing for transplantation between individuals), their lack of ethical controversy, their potential to differentiate into tissue-specific cell types, and their ability to promote blood vessel growth. The cells can seek out and repair damaged tissue and treat inflammation resulting from cardiovascular disease, brain and spinal cord injury, cartilage and bone injury, Crohn's disease, and other conditions. Given the systemic nature of many … Continue reading
