Scientists drill down to genetic root of prostate tumor development

IMAGE:This infographic shows how prostate cancer spreads. view more Credit: Cancer Research UK Scientists have revealed the root of prostate cancers in individual men, discovering that despite huge genetic variety between tumours they also share common gene faults - insight that could offer new treatment hopes, according to research published in Nature today (Wednesday). In a landmark paper, Cancer Research UK funded scientists alongside an international team of researchers read all of the DNA in tumour samples from 10 men with prostate cancer. This allowed them to map a 'family tree' of the changes happening at a genetic level as the disease spreads, forms new tumours, and becomes resistant to treatment. They also revealed more detail about how prostate cancer spreads, showing that the group of cells that first spread from the prostate carry on travelling around the body, forming more secondary tumours. The research is part of the International Cancer Genome Consortium (ICGC) - a global project using the latest gene-sequencing technology to reveal the genetic changes driving the disease. Premature aging of stem cell telomeres, not inflammation, linked to emphysema

Lung diseases like emphysema and pulmonary fibrosis are common among people with malfunctioning telomeres, the "caps" or ends of chromosomes. Now, researchers from Johns Hopkins say they have discovered what goes wrong and why. Mary Armanios, M.D., an associate professor of oncology at the Johns Hopkins University School of Medicine., and her colleagues report that some stem cells vital to lung cell oxygenation undergo premature aging -- and stop dividing and proliferating -- when their telomeres are defective. The stem cells are those in the alveoli, the tiny air exchange sacs where blood takes up oxygen. In studies of these isolated stem cells and in mice, Armanios' team discovered that dormant or senescent stem cells send out signals that recruit immune molecules to the lungs and cause the severe inflammation that is also a hallmark of emphysema and related lung diseases. Until now, Armanios says, researchers and clinicians have thought that "inflammation alone is what drives these lung diseases and have based therapy on anti-inflammatory drugs for the last 30 years." Catabasis raising $20.4M; modulates multiple disease pathways for Duchenne muscular dystrophy, ALS

Multiple biological pathways go awry to cause disease. Cambridge startup Catabasis is developing a platform to simultaneously modulate multiple targets in a disease pathway, with a focus on diseases like Duchenne muscular dystrophy and ALS The startupsin the midst of a $20.4 million fundraise, according to a recent SEC filing. So far, its raised about $12.4 million in this round. The company says its platform can simultaneously modulate a number of biological targets within several related disease pathways. These are small chemicals designed to join two separate bioactives into a single conjugate molecule, Catabasis says. The platforms called Safely Metabolized And Rationally Targeted, shortened to SMART. The MIT Technology Review wrote up a nice explainer of Catabasis technology two years ago: Catabasis has found a way around the challenge of identifying a single active molecule that can hit multiple pathways effectively: using a chemical linker to bring together two active molecules.
